OPTIMIZATION, DEVELOPMENT AND EVALUATION OF REPAGLINIDE CONTROLLED RELEASE GASTRO-RETENTIVE FLOATING TABLET USING CENTRAL COMPOSITE DESIGN

Objective: The recent study's objective was to optimize and formulate a controlled-release gastro-retentive floating tablet of RG using central composite design, which provides continuous release Repaglinide for up 24 h. Methods: (RG-GRF Tablet) prepared by direct compression method. optimization carried out three-factor three-level Central Composite design. amount Eudragit RSPO (A), HPMC K-100M (B) Sodium bicarbonate (C) were selected as independent variables the Cumulative % drug in 1.5 h (DR1.5), 8 (DR8), (DR24) Floating lag time (FLT) used dependent variables. Results: CCD analysis results shows that predicted experimental values optimized formulation found be almost similar. Optimized amounts RSPO, K-100M, NaHCO3 14.351 mg, 44.438 10 respectively, with highest possible desirability value 0.898. at preparation conditions DR1.5 is 30.68%, DR8 64.90%, DR24 96.54%, FLT 4.41 min. data from closely matched Korsmeyer-Peppas model vitro studies indicated RG-GRF Tablet continuously releases controlled manner. Conclusion: Current research concludes Tablets provide h, derived design can forecasting DR1.5, well. also made more bioavailable extending gastric residence time.
